Disease ID:DOID:684D006528
Disease Name:hepatocellular carcinomaCarcinoma, Hepatocellular
Category:Disease OntologyMeSH
Type:Neoplasms
Define:A liver carcinoma that has_material_basis_in undifferentiated hepatocytes and located_in the liver.A primary malignant neoplasm of epithelial liver cells. It ranges from a well-differentiated tumor with EPITHELIAL CELLS indistinguishable from normal HEPATOCYTES to a poorly differentiated neoplasm. The cells may be uniform or markedly pleomorphic, or form GIANT CELLS. Several classification schemes have been suggested.

Reference

[1] PubMed ID:34093813
Disease Name:Carcinoma, Hepatocellular
Sample:HCC tissues and cell lines
Dysfunction Pattern:Regulation(CREB/PFKFB4 )
Validated Method:CCK8//qRT-PCR//Western Blot
Description: Here, we disclosed that the FIRRE level was frequently increased in HCC compared to nontumor tissues. Compared with normal liver cells, we also confirmed the upregulated level of FIRRE in HCC cells.Mechanistically, modulating FIRRE level did not affect the stability of PFKFB4 mRNA. FIRRE was mainly distributed in HCC cells' nucleus and promoted PFKFB4 transcription and expression via cAMP-responsive element-binding protein (CREB). PFKFB4 could abolish the effects of FIRRE knockdown on HCC cell proliferation and glycolysis.
Causality:Yes
Causal Description:Functionally, FIRRE knockdown repressed the proliferation and glycolysis of HCCLM3 cells.Overexpression of FIRRE strengthened Huh7 cell proliferation and glycolysis. Notably, FIRRE positively regulated the glycolic enzyme 6-phosphofructo-2-kinase/fructose-2,6-biphosphatase 4 (PFKFB4) expression in HCC cells.
Clinical-realted Application:Notably, the FIRRE high expression was related to malignant clinical features, including advanced TNM stage and tumor size ≥5 cm, and conferred to worse survival of HCC.

[2] PubMed ID:35988459
Disease Name:Carcinoma, Hepatocellular
Sample:cell lines and tissue samples
Dysfunction Pattern:Interaction(MBNL3/PXN )
Validated Method:qRT-PCR
Description:In this study, we found that FIRRE, a recently identified cancer-associated lncRNA located on chromosome X, is highly expressed in HCC cell lines and tissue samples, and its expression is positively correlated with poor HCC prognosis. As for the potential mechanism, FIRRE specifically binds to the splicing factor MBNL3 to affect the expression of PXN to regulate the pathological characteristics of HCC cells.
Causality:Yes
Causal Description: In vitro and in vivo functional analyses showed that FIRRE could promote the proliferation, migration, and invasion of HCC.
Clinical-realted Application:In this study, we found that FIRRE, a recently identified cancer-associated lncRNA located on chromosome X, is highly expressed in HCC cell lines and tissue samples, and its expression is positively correlated with poor HCC prognosis.
