Disease ID:DOID:1909D008545
Disease Name:melanomaMelanoma
Category:Disease OntologyMeSH
Type:Neoplasms
Define:A cell type cancer that has_material_basis_in abnormally proliferating cells derives_from melanocytes which are found in skin, the bowel and the eye.A malignant neoplasm derived from cells that are capable of forming melanin, which may occur in the skin of any part of the body, in the eye, or, rarely, in the mucous membranes of the genitalia, anus, oral cavity, or other sites. It occurs mostly in adults and may originate de novo or from a pigmented nevus or malignant lentigo. Melanomas frequently metastasize widely, and the regional lymph nodes, liver, lungs, and brain are likely to be involved. The incidence of malignant skin melanomas is rising rapidly in all parts of the world. (Stedman, 25th ed; from Rook et al., Textbook of Dermatology, 4th ed, p2445)

[1] PubMed ID:31838468
Disease Name:Melanoma
Sample:melanoma tissues and cells
Dysfunction Pattern:Regulation[Wnt/β-Catenin Signaling Pathway]
Validated Method:Western Blot//Flow Cytometry//qRT-PCR//Luciferase Report Assay//Colony Formation Assay//Invasion Assay//Transwell Assay
Description:The expression of CASC15 and β-catenin was upregulated in melanoma tissues and cells. Our study demonstrated that CASC15 promoted the proliferation, migration, and invasion of melanoma cells via activating Wnt/β-catenin signaling pathway, implying that CASC15 might be a potential therapeutic target and prognostic biomarker for melanoma.
Causality:Yes
Causal Description: Furthermore, inhibition of CASC15 decreased proliferation and increased apoptosis of melanoma cells by downregulating Survivin and Bcl-2 and upregulating Bax in A375 and SK-MEL-28 cells. Silencing of CASC15 inhibited migration and invasion of melanoma cells by repressing EMT process.

[2] PubMed ID:30013768
Disease Name:Melanoma
Sample:melanoma tissues
Dysfunction Pattern:interaction[PDCD4]
Validated Method:Western Blot//CCK8//qRT-PCR//RIP//Luciferase Report Assay//Cell Apoptosis Assay//cell invasion assay//Cell Cycle Assay
Description:We found that CASC15 expression was up-regulated in melanoma tissues and associated with advanced pathological stages. CASC15 acts as an oncogene by negatively regulating PDCD4 expression via recruiting EZH2 and subsequently increasing H3K27me3 level.
Causality:Yes
Causal Description: Function experiments displayed that CASC15 knockdown hindered proliferation, facilitated apoptosis and suppressed invasion, while CASC15 overexpression facilitated proliferation and invasion in melanoma cells.
Clinical-realted Application:
