Disease Information

 Disease OntologyMeSH
Disease ID:DOID:0050827D012214
Disease Name:rheumatic heart diseaseRheumatic Heart Disease
Category:Disease OntologyMeSH
Type:Infections
Define:A heart valve disease that is characterized by repeated inflammation with fibrinous repair caused by an autoimmune reaction to Group A beta-hemolytic streptococci (GAS) that results in valvular damage. The cardinal anatomic changes of the valve include leaflet thickening, commissural fusion, and shortening and thickening of the tendinous cords.Cardiac manifestation of systemic rheumatological conditions, such as RHEUMATIC FEVER. Rheumatic heart disease can involve any part the heart, most often the HEART VALVES and the ENDOCARDIUM.

[1] PubMed ID:34695392
Disease Name:Rheumatic Heart Disease
Sample:mitral valve tissues from three mitral stenosis patients with RHD
Dysfunction Pattern:Regulation
Validated Method:In Vivo Experiment//Western Blot//qRT-PCR//RNA-seq//Luciferase Report Assay//Bioinformatics Analysis
Description:The presence of hypomethylated CpG islands at promoter and 5' terminal of PRKCA was observed in RHD accompanied with highly expressed PRKCA and down-regulated antisense long non-coding RNA (lncRNA) PRKCA-AS1 compared to health control. Additionally, PRKCA-AS1 was further determined to bind with promoter of PRKCA via 5' terminal and interact with DNMT1 via 3' terminal.

[2] PubMed ID:34482802
Disease Name:Rheumatic Heart Disease
Sample:three mitral stenosis patients with RHD
Dysfunction Pattern:Expression(highly expressed)
Validated Method:FISH//ELISA//qPCR//Western Blot
Description:The presence of hypomethylated CpG islands at promoter and 5' terminal of PRKCA was observed in RHD accompanied with highly expressed PRKCA and down-regulated antisense long non-coding RNA (lncRNA) PRKCA-AS1 compared to health control. Furthermore, the enrichments of DNMT1/3A/3B on PRKCA were detected by ChIP-qPCR assay in vivo and in human cardiomyocyte AC16 and RL-14 cells exposed to TNF-α in vitro, and both demonstrated that DNMT1 substantially contributed to DNA methylation.
